Background: Edible films incorporating natural bioactive compounds offer a novel approach to environmental preservation and extending food shelf life. This study aimed to investigate the effect of different extraction solvents (water, methanol, and ethanol) on the total phenolic content and antioxidant activity of pomegranate peel extract. Subsequently, an active film based on hydroxypropyl methylcellulose (HPMC) was developed and evaluated.
Methods: The total phenolic content of the extracts was determined using the Folin-Ciocalteu method, and antioxidant activity was evaluated via the DPPH assay. The oxidative stability of soybean oil packaged with HPMC films containing three concentrations (200, 400, and 600 ppm) of the extracts was assessed over 60 days by measuring the peroxide value. Mechanical properties were analyzed using tensile testing, and antibacterial activity against Staphylococcus aureus, Salmonella Typhimurium, and Escherichia coli was examined using the disc diffusion method. Data were analyzed by ANOVA followed by Duncan's multiple range test (P < 0.05).
Findings: The methanolic extract exhibited the highest total phenolic content and antioxidant activity compared to the ethanolic and aqueous extracts. The film containing the methanolic extract demonstrated the greatest improvement in the oxidative stability of soybean oil, with the efficacy of the films in slowing oxidation increasing over time. Additionally, the film with the methanolic extract was the most effective treatment in inhibiting bacterial growth, particularly against Staphylococcus aureus. The incorporation of the extracts generally enhanced the flexibility of the HPMC films.
Conclusion: The hydroxypropyl methylcellulose film containing methanolic pomegranate peel extract, serving as active packaging with antioxidant and antimicrobial properties, has significant potential for application in the food industry.
